Many people shopping for discounted second-hand items online use the eBay or Amazon websites. Taking that into consideration, you may want to set up your online store to receive traffic from those websites so that you can take advantage of those buyers.You can set up an eBay Store for less than $20/month.
You can set up a storefront on Amazon.com also.
If you want to set up an online store separate from eBay and Amazon, you can use an online store program like Prostores.com or BigCommerce.com which include pretty much all the hosting, design and shopping cart tools you'll need.

More and more people are bargain shopping these days, so selling discounted items I think is the way to go. However, it's going to be pretty hard to compete with sites like eBay - unless you have an eBay store and sell through eBay.
If you have your own online thrift store, I think you'd do better to specialize in a particular area or type of product. Just my 2 cents.
